Acute viral infection results in a PD-1-dependent loss of anti-tumor CD8+ T cell responses: implications for tumor immunotherapy
نویسندگان
چکیده
Methods To test this hypothesis, we used the A/H1N1/PR8 mouse model of influenza infection (10,000 pfu, via intranasal administration) prior to or after challenge with B16 melanoma (12,000 120,000 cells via intradermal injection in the right flank). Kaplan-Meier curves were used to present animal survival and tumor incidence data and log rank test was used to compare such curves. All statistical analyses were performed using Prism software (v4.0, GraphPad software), and a p value of less than 0.05 was considered to denote statistically significant differences among compared variables.

Producing Soluble Human Programmed Cell Death Protein-1: A Natural supporter for CD4+T cell Cytotoxicity and Tumor Cells Apoptosis
Background: Programmed cell death protein-1 (PD-1)/PD-L1 pathway is one of the immune checkpoint pathways involved in regulation of the immune responses and suppression of anti-tumor defense. PD-1/B7-H1-blocking antibodies improve immune responses such as cytotoxic activity of CD8+/CD4+T cells and also increase mortality of tumor cells; however their use is accompanied by adverse effects in pat...
متن کاملPD-1 blockade reverses viral infection-induced loss of anti-tumor CD8+ T cell responses
Background Emerging epidemiologic studies describe an increased prevalence of tumors in patients with non-oncogenic chronic viral disease (e.g., non-AIDS-defining cancers in HIV and non-hepatic cancers in HCV). However, there is a lack of basic understanding by what mechanism infections result in increased unrelated cancer formation (i.e., of cancers in tissues not associated with the infection...

متن کاملListeria Monocytogenes Activated Dendritic Cell Based Vaccine for Prevention of Experimental Tumor in Mice
Background: The use of dendritic cells (DCs) as a cellular adjuvant provides a promis-ing approach in immunotherapy of cancer. It has been demonstrated that Listeria mono-cytogenes activated DCs pulsed ex vivo with tumor antigens trigger a systemic Th1-biased specific immune response and a single dose of this vaccine will cause a consider-able anti tumor immunity. Objective: The present study w...
متن کاملNew approaches in cancer immunotherapy: review article
Cancer immunotherapy refers to any intervention that leverages the immune system to eliminate a malignancy. Successful cancer immunotherapies generate an anti-cancer response that is systemic, specific, and durable and overcome to the primary limitations of traditional cancer treatment modalities. In this review paper, the effective methods in immune system to treat cancer, such as immunosuppre...
